  • Orforglipron, a Daily GLP-1 Pill, Cut Weight in Adults With Diabetes

    The GLP-1 drugs that reshaped weight loss have mostly come as weekly injections. A pill would change who can reach them. In a trial of 1,613 adults who had both obesity and type 2 diabetes, the once-daily tablet orforglipron cut body weight by up to 9.6% over 72 weeks, against 2.5% for a placebo pill, while also pulling down blood sugar.1 No needle, no refrigeration, no timing it around food or water.

    A Pill That Acts Like the Injections

    Orforglipron mimics GLP-1, a gut hormone that quiets appetite and steadies blood sugar after eating. Semaglutide and tirzepatide do the same, but they’re injected, and the one oral GLP-1 already sold must be taken on an empty stomach with strict rules. Orforglipron is a small molecule, sturdy enough to swallow as an ordinary tablet. That’s why regulators’ approval of it in April 2026 drew notice: for people with diabetes who won’t or can’t inject, an equally simple pill widens the door.

    Three Doses, Randomly Assigned

    The trial spanned adults with obesity or overweight plus type 2 diabetes, average starting blood sugar around 8%. A computer sorted them into one of three orforglipron doses or a placebo, and neither they nor their doctors knew which. Over 72 weeks the results tracked the dose. Weight fell 5.1% on the lowest dose and 9.6% on the highest, versus 2.5% on placebo.1 HbA1c, the three-month blood sugar average, dropped up to 1.8 percentage points against 0.1 on placebo, and most people on the drug finished under the 6.5% mark that signals good control. A larger companion trial in people without diabetes found weight loss reaching 11.2%.2

    The Familiar GLP-1 Trade-Off

    The side effects are the ones this drug class is known for: nausea, vomiting, diarrhea and constipation, mostly mild to moderate. They pushed 6% to 10% of people off the drug, compared with 4% on placebo.1 One honest limit: weight loss on a pill you swallow tends to trail the strongest injections, so a tablet is not automatically the most powerful option. It’s the most reachable one. For a person weighing whether to start any of these medicines, the choice between a shot and a pill is now a real choice rather than a foregone one.
